There's an Australian film called "Love and Other Catastrophes" which I'd recommend, although it'd probably be hard to get ahold of for people outside of Australia.
Also you should watch "But I'm A Cheerleader", that's a cute and funny movie.
How about 'The Incredibly True Adventure of Two Girls in Love.' Not the best movie ever, but it does meat the cute/chick criteria. Also 'Saving Face' is great and 'Love and Other Catastrophes' is really cute.
I dunno, Gia might not be the happiest film. Brilliant, but yeah, not very happy and cute. Nina's Heavenly Delights, as someone mentioned, is pure feel good. Or just watch IMAY again, it bears re-watching :D
I agree with the people who recommended But I'm a Cheerleader. It's probably the Cutest, Funniest, Happiest, lesbian movie I have seen.
I would also add Better Than Chocolate. It's a little over the top in places (well, so is But I'm a Cheerleader, but that is on purpose) and not the most well written, but still very cute, and pretty funny.
There's Girl Play and Mango Kiss for the lighthearted angle. High Art for less of a happy ending.... Eulogy isn't exactly a Lesbian film but Famke Jansson and Kelly Preston are a very cute couple in it. Debra Winger and Glenne Headly have a steamy car scene. Plus the movie is wicked funny.
... the short film collection "Watching You." There's some fun stuff in there, nobody dies, nobody runs around looking for sperm, nobody goes crazy ... did I mention nobody dies?
Oh, one of the shorts is Michele Paradise's "10 Rules ..." which is funny, sweet, though kinda familar if you've seen "Exes and Ohs."
This Finnish drama (with English subtitles) is not exactly a romcom, but it has definite moments of humor, and nobody goes crazy or dies. Fair warning: if you are hetero-adverse or hostile toward bi's, this is not for you, but if you are a fair-minded grown-up it is a good movie.
"Naissance Des Pieuvres" (or "Water Lilies" in english). It's a coming of age film, very honest and vey subtle. The director is a lesbian and she did an impressive job considering that it was her first feature film (she also wrote the script).
Definately not cute nor happy, but i strongly recommend "Mulholland Dr.", i admit it's not an easy film to watch, but it's probably my favorite depiction in a film of two women falling in love with each other (and the first sex scene is amazing!).
Prettymuch every movie named here, plus some more can be found at the website for wolfe films. Granted, it's more for ordering dvds, but it's good place to find titles to look for in your local Hollywood Video.
Not necessarily a good movie, but definitely a cute, happy movie. The best parts are watching the two women watching/looking at each other. And the confrontation with the mother is fabulous!
(and some have already been mentioned here), that it might be easier to suggest the few movies that you might want to avoid.. "Lost and Delirious" and "High Art" will keep you depressed for days . "Bound" might give you an anxiety attack. "Aimee and Jaguar" (though it does have some happy towards the end) won't leave you laughing. These are all good movies though.
I LOVE the movie Bound! I've seen it over 100 times. While it may not be the cutest movie, it is funny at some points, but there are two very sexy sex scenes!
I also recommend D.E.B.S.(cute, funny, and sexy...I LOVE Jordana Brewster!) Lost & Delirious(cute, but sad), Loving Annabelle(cute, and funny, but also sad), and Saving Face(cute and funny...I just think they should have focused the camera on Wil and Vivian more than they did).
Not a big fan of Go Fish, The Girl, or When the Night is Falling, but to each her own, I guess. =)
I definately reccomend show me love. It's a great film and the girls are very cute(In a non-sexual way. They are too young for me to be attracted to them. Well, Rebecca was 17 but looked about 14!). D.E.B.S is nice and light hearted. Imagine me and You, But I'm a Cheerleader, Saving Face are others I would reccomend for that type of film.
I still adore "Clarie Of The Moon" directed by Nicole Conn, a story of two writers from different universes at an all women's writing retreat. There is also a really cute/sweet kind of cross between Thelma & Louise and Fried Green Tomatoes flick called "Leaving Normal" with Christine Lahti and Meg Tilly. Also, "Puccini For Beginners" was directed by writer/director Maria Maggenti who also directed the other above mentioned romantic comedy "The Incredibly True Adventure of Two Girls in Love". "Bound" will always be one of my all time favorites (Gina Gershon and Jennifer Tilly, really, need we say more?). "Tipping the Velvet" based on a book written by Sarah Waters and starring Keeley Hawes. Finally, not to date myself prematurely or anything, the first lesbian/bi themed movie I saw was "Personal Best"; if you can tolerate Mariel Hemingway's "whining" character, it is pretty good.
You'll definitely want to watch "When Night is Falling" as one or two people recommended above. It's cute and charming and had a happy ending. I loved it.
I'm surprised no one has mentioned this one yet (unless I missed it). One of the cutest lesbian flicks I've seen, and great for 20-somethings just coming out. Also, Imagine Me and You is great, in the lighthearted Brit-Com tradition.
My Summer of Love
Yeah, not really. Not exactly cute or happy but still very good. Mmmm Saving Face?

Cute and Happy?
I read somewhere someone suggested Loving Annabelle, I dont think that falls into the cute and Happy category.
If you are looking for the non cute and happy Id go with the following:
Loving Annabelle
Gia
Lost and Delirious
Love and Suicide
But as for cute and happy Imagine Me and You, Saving Face and D.E.B.S. take the cake for me. Maybe Puccinni for Beginners?
And some more....
Bound
Among a long list I would recommend "BOUND" , still big fan & it is having a happy ending.
